Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_001370259.2(MEN1):c.245A>C (p.Asp82Ala), citing Ambry Variant Classification Scheme 2023: The p.D82A variant (also known as c.245A>C), located in coding exon 1 of the MEN1 gene, results from an A to C substitution at nucleotide position 245. The aspartic acid at codon 82 is replaced by alanine, an amino acid with dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
