Uncertain significance — the classification assigned by Ambry Genetics to NM_001365951.3(KIF1B):c.2590T>A (p.Ser864Thr), citing Ambry Variant Classification Scheme 2023: The p.S818T variant (also known as c.2452T>A), located in coding exon 23 of the KIF1B gene, results from a T to A substitution at nucleotide position 2452. The serine at codon 818 is replaced by threonine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. The evidence for this gene-disease relationship is limited; therefore, the clinical significance of this alteration is unclear.
