Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001184.4(ATR):c.2441A>G (p.Glu814Gly), citing Ambry Variant Classification Scheme 2023: The p.E814G variant (also known as c.2441A>G), located in coding exon 11 of the ATR gene, results from an A to G substitution at nucleotide position 2441. The glutamic acid at codon 814 is replaced by glycine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
