NM_001430.5(EPAS1):c.2436C>A (p.Ser812Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S812R variant (also known as c.2436C>A), located in coding exon 15 of the EPAS1 gene, results from a C to A substitution at nucleotide position 2436. The serine at codon 812 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
