Submitter rationale: The c.2435A>G (p.E812G) alteration is located in exon 29 (coding exon 29) of the COL5A1 gene. This alteration results from an A to G substitution at nucleotide position 2435, causing the glutamic acid (E) at amino acid position 812 to be replaced by a glycine (G). This variant was not reported in population-based cohorts in the Genome Aggregation Database (gnomAD). Another alteration at the same codon, c.2436A>T (p.E812D), has been detected in one individual with doughy skin, atrophic scarring, hypermobility, mild prognathism, easy bruising, and dental anomalies (Ritelli, 2013).
