Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000051.4(ATM):c.2431C>G (p.Leu811Val), citing Ambry Variant Classification Scheme 2023: The c.2431C>G variant (also known as p.L811V), located in coding exon 15 of the ATM gene, results from a C to G substitution at nucleotide position 2431. The leucine at codon 811 is replaced by valine, an amino acid with highly similar properties. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site and will result in the creation or strengthening of a novel splice donor site. RNA studies have demonstrated that this alteration results in a transcript predicted to lead to a protein with an in-frame deletion of 12 amino acids; however, the exact functional impact of the deleted amino acids are unknown at this time (Ambry internal data). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
