Uncertain significance — the classification assigned by Ambry Genetics to NM_001166108.2(PALLD):c.2480G>C (p.Trp827Ser), citing Ambry Variant Classification Scheme 2023: The p.W810S variant (also known as c.2429G>C), located in coding exon 13 of the PALLD gene, results from a G to C substitution at nucleotide position 2429. The tryptophan at codon 810 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
