Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_014795.4(ZEB2):c.2425G>A (p.Glu809Lys), citing Ambry Variant Classification Scheme 2023: The p.E809K variant (also known as c.2425G>A), located in coding exon 7 of the ZEB2 gene, results from a G to A substitution at nucleotide position 2425. The glutamic acid at codon 809 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
