NM_001430.5(EPAS1):c.2411A>T (p.Tyr804Phe) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Y804F variant (also known as c.2411A>T), located in coding exon 15 of the EPAS1 gene, results from an A to T substitution at nucleotide position 2411. The tyrosine at codon 804 is replaced by phenylalanine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
