NM_017802.4(DNAAF5):c.2379G>T (p.Glu793Asp) was classified as Uncertain significance for Primary ciliary dyskinesia by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E793D variant (also known as c.2379G>T), located in coding exon 12 of the DNAAF5 gene, results from a G to T substitution at nucleotide position 2379. The glutamic acid at codon 793 is replaced by aspartic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
