NM_001378454.1(ALMS1):c.10952A>T (p.His3651Leu) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H3652L variant (also known as c.10955A>T), located in coding exon 16 of the ALMS1 gene, results from an A to T substitution at nucleotide position 10955. The histidine at codon 3652 is replaced by leucine, an amino acid with similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
