Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_177438.3(DICER1):c.235A>G (p.Lys79Glu), citing Ambry Variant Classification Scheme 2023: The p.K79E variant (also known as c.235A>G), located in coding exon 2 of the DICER1 gene, results from an A to G substitution at nucleotide position 235. The lysine at codon 79 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
