NM_001267550.2(TTN):c.50721_50723dup (p.Lys16908_Asp16909insLys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023. This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 50721 through coding-DNA position 50723, duplicating 3 bases. Submitter rationale: The c.23526_23528dupAAA variant (also known as p.K7843dup), located in coding exon 96 of the TTN gene, results from an in-frame duplication of AAA at nucleotide positions 23526 to 23528. This results in the duplication of an extra lysine residue between codons 7843 and 7844. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
