Submitter rationale: The p.E780K variant (also known as c.2338G>A), located in coding exon 15 of the SMARCA4 gene, results from a G to A substitution at nucleotide position 2338. The glutamic acid at codon 780 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. This missense alteration is located in a region that has a low rate of benign missense variation (Lek M et al.
