NM_001374736.1(DST):c.17287A>G (p.Ile5763Val) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I3644V variant (also known as c.10930A>G), located in coding exon 60 of the DST gene, results from an A to G substitution at nucleotide position 10930. The isoleucine at codon 3644 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and threonine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
