NM_000245.4(MET):c.2282C>T (p.Thr761Ile) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.T779I variant (also known as c.2336C>T), located in coding exon 9 of the MET gene, results from a C to T substitution at nucleotide position 2336. The threonine at codon 779 is replaced by isoleucine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
