NM_001365951.3(KIF1B):c.2449G>A (p.Asp817Asn) was classified as Uncertain significance by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D771N variant (also known as c.2311G>A), located in coding exon 22 of the KIF1B gene, results from a G to A substitution at nucleotide position 2311. The aspartic acid at codon 771 is replaced by asparagine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
