NM_000057.4(BLM):c.2305A>C (p.Lys769Gln) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K769Q variant (also known as c.2305A>C), located in coding exon 9 of the BLM gene, results from an A to C substitution at nucleotide position 2305. The lysine at codon 769 is replaced by glutamine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
