Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_020975.6(RET):c.2269G>A (p.Val757Met), citing Ambry Variant Classification Scheme 2023: The p.V757M variant (also known as c.2269G>A), located in coding exon 12 of the RET gene, results from a G to A substitution at nucleotide position 2269. The valine at codon 757 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
