Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001999.4(FBN2):c.2257C>T (p.His753Tyr), citing Ambry Variant Classification Scheme 2023: The p.H753Y variant (also known as c.2257C>T), located in coding exon 17 of the FBN2 gene, results from a C to T substitution at nucleotide position 2257. The histidine at codon 753 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
