Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_001369.3(DNAH5):c.2254A>G (p.Met752Val), citing Ambry Variant Classification Scheme 2023: The p.M752V variant (also known as c.2254A>G), located in coding exon 15 of the DNAH5 gene, results from an A to G substitution at nucleotide position 2254. The methionine at codon 752 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
