Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_004064.5(CDKN1B):c.224A>G (p.Glu75Gly), citing Ambry Variant Classification Scheme 2023: The p.E75G variant (also known as c.224A>G), located in coding exon 1 of the CDKN1B gene, results from an A to G substitution at nucleotide position 224. The glutamic acid at codon 75 is replaced by glycine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
