NM_000249.4(MLH1):c.2246T>G (p.Leu749Arg) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L749R variant (also known as c.2246T>G), located in coding exon 19 of the MLH1 gene, results from a T to G substitution at nucleotide position 2246. The leucine at codon 749 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species.
