NM_000202.8(IDS):c.223C>G (p.Gln75Glu) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q75E variant (also known as c.223C>G), located in coding exon 2 of the IDS gene, results from a C to G substitution at nucleotide position 223. The glutamine at codon 75 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
