NM_001184.4(ATR):c.2230A>T (p.Asn744Tyr) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.N744Y variant (also known as c.2230A>T), located in coding exon 10 of the ATR gene, results from an A to T substitution at nucleotide position 2230. The asparagine at codon 744 is replaced by tyrosine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
