Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.1087A>G (p.Lys363Glu), citing Ambry Variant Classification Scheme 2023: The p.K363E variant (also known as c.1087A>G), located in coding exon 10 of the RAD54L gene, results from an A to G substitution at nucleotide position 1087. The lysine at codon 363 is replaced by glutamic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
