Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_000033.4(ABCD1):c.221G>A (p.Arg74Gln), citing Ambry Variant Classification Scheme 2023: The p.R74Q variant (also known as c.221G>A), located in coding exon 1 of the ABCD1 gene, results from a G to A substitution at nucleotide position 221. The arginine at codon 74 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
