Uncertain significance — the classification assigned by Ambry Genetics to NM_003579.4(RAD54L):c.2213G>T (p.Arg738Leu), citing Ambry Variant Classification Scheme 2023: The p.R738L variant (also known as c.2213G>T), located in coding exon 18 of the RAD54L gene, results from a G to T substitution at nucleotide position 2213. The arginine at codon 738 is replaced by leucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
