Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_003238.6(TGFB2):c.1086G>A (p.Arg362=), citing Ambry Variant Classification Scheme 2023: The c.1086G>A variant (also known as p.R362R), located in coding exon 6 of the TGFB2 gene, results from a G to A substitution at nucleotide position 1086. This nucleotide substitution does not change the arginine at codon 362. However, this change occurs in the last base pair of coding exon 6, which makes it likely to have some effect on normal mRNA splicing. This nucleotide position is not well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will not have any significant effect on splicing. Based on the available evidence, the clinical significance of this variant remains unclear.
