NM_000388.4(CASR):c.2205G>C (p.Gln735His) was classified as Uncertain significance for Nephrolithiasis/nephrocalcinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.Q735H variant (also known as c.2205G>C), located in coding exon 6 of the CASR gene, results from a G to C substitution at nucleotide position 2205. The glutamine at codon 735 is replaced by histidine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
