Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_001110556.2(FLNA):c.2203T>C (p.Tyr735His), citing Ambry Variant Classification Scheme 2023: The p.Y735H variant (also known as c.2203T>C), located in coding exon 14 of the FLNA gene, results from a T to C substitution at nucleotide position 2203. The tyrosine at codon 735 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
