Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.11956A>G (p.Ile3986Val), citing Ambry Variant Classification Scheme 2023: The p.I3623V variant (also known as c.10867A>G), located in coding exon 44 of the TTN gene, results from an A to G substitution at nucleotide position 10867. The isoleucine at codon 3623 is replaced by valine, an amino acid with highly similar properties. This amino acid position is not well conserved in available vertebrate species, and valine is the reference amino acid in other vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
