Pathogenic for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000249.4(MLH1):c.1086_1092dup (p.Ser365fs), citing Ambry Variant Classification Scheme 2023: The c.1086_1092dupCACAACA pathogenic mutation, located in coding exon 12 of the MLH1 gene, results from a duplication of CACAACA at nucleotide position 1086, causing a translational frameshift with a predicted alternate stop codon (p.S365Hfs*13). This alteration is expected to result in loss of function by premature protein truncation or nonsense-mediated mRNA decay. As such, this alteration is interpreted as a disease-causing mutation.
