Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_017802.4(DNAAF5):c.218G>C (p.Trp73Ser), citing Ambry Variant Classification Scheme 2023: The p.W73S variant (also known as c.218G>C), located in coding exon 1 of the DNAAF5 gene, results from a G to C substitution at nucleotide position 218. The tryptophan at codon 73 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
