This variant lies in the NBN gene (transcript NM_002485.5) at the canonical splice donor site of the intron immediately after coding-DNA position 2184 through 5 bases into the intron immediately after coding-DNA position 2184, deleting this region. Submitter rationale: The c.2184+1_2184+5delGTTAG intronic variant, located in intron 14 of the NBN gene, results from a deletion of 5 nucleotides within intron 14 of the NBN gene. This nucleotide position is well conserved in available vertebrate species. In silico splice site analysis predicts that this alteration will weaken the native splice donor site. The resulting transcript is predicted to be in-frame and is not expected to trigger nonsense-mediated mRNAdecay; however, direct evidence is unavailable.
