NM_007078.3(LDB3):c.2180T>C (p.Leu727Ser) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L727S variant (also known as c.2180T>C), located in coding exon 13 of the LDB3 gene, results from a T to C substitution at nucleotide position 2180. The leucine at codon 727 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
