Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_017636.4(TRPM4):c.2173G>C (p.Asp725His), citing Ambry Variant Classification Scheme 2023: The p.D725H variant (also known as c.2173G>C), located in coding exon 16 of the TRPM4 gene, results from a G to C substitution at nucleotide position 2173. The aspartic acid at codon 725 is replaced by histidine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
