Uncertain significance — the classification assigned by Ambry Genetics to NM_013266.4(CTNNA3):c.2162G>A (p.Gly721Asp), citing Ambry Variant Classification Scheme 2023: The p.G721D variant (also known as c.2162G>A), located in coding exon 15 of the CTNNA3 gene, results from a G to A substitution at nucleotide position 2162. The glycine at codon 721 is replaced by aspartic acid, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
