NM_004360.5(CDH1):c.215A>T (p.Asp72Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.D72V variant (also known as c.215A>T), located in coding exon 3 of the CDH1 gene, results from an A to T substitution at nucleotide position 215. The aspartic acid at codon 72 is replaced by valine, an amino acid with highly dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
