NM_000264.5(PTCH1):c.214G>A (p.Gly72Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.G72S variant (also known as c.214G>A), located in coding exon 2 of the PTCH1 gene, results from a G to A substitution at nucleotide position 214. The glycine at codon 72 is replaced by serine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
