NM_001349253.2(SCN11A):c.1081T>C (p.Trp361Arg) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.W361R variant (also known as c.1081T>C), located in coding exon 8 of the SCN11A gene, results from a T to C substitution at nucleotide position 1081. The tryptophan at codon 361 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
