NM_000548.5(TSC2):c.213G>C (p.Lys71Asn) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.K71N variant (also known as c.213G>C), located in coding exon 2 of the TSC2 gene, results from a G to C substitution at nucleotide position 213. The lysine at codon 71 is replaced by asparagine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
