NM_000251.3(MSH2):c.2135T>C (p.Val712Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.V712A variant (also known as c.2135T>C), located in coding exon 13 of the MSH2 gene, results from a T to C substitution at nucleotide position 2135. The valine at codon 712 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
