Uncertain significance for Inborn genetic diseases — the classification assigned by Ambry Genetics to NM_001430.5(EPAS1):c.212C>T (p.Ser71Phe), citing Ambry Variant Classification Scheme 2023: The p.S71F variant (also known as c.212C>T), located in coding exon 2 of the EPAS1 gene, results from a C to T substitution at nucleotide position 212. The serine at codon 71 is replaced by phenylalanine, an amino acid with highly dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
