Submitter rationale: The c.2114_2120delCCAGGGAinsG variant (also known as p.A705_D707delinsG), located in coding exon 14 of the LDLR gene, results from an in-frame deletion of 7 nucleotides and insertion of one nucleotide at nucleotide positions 2114 to 2120. This results in the substitution of the alanine, arginine, and aspartic acid residues for a glycine residue at codon 705. These amino acid positions are not well conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis (Choi Y et al. PLoS ONE. 2012; 7(10):e46688). Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
