Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001267550.2(TTN):c.48295C>T (p.Arg16099Trp), citing Ambry Variant Classification Scheme 2023: The p.R7034W variant (also known as c.21100C>T), located in coding exon 84 of the TTN gene, results from a C to T substitution at nucleotide position 21100. The arginine at codon 7034 is replaced by tryptophan, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
