Submitter rationale: The c.2109_2122del14 pathogenic mutation, located in coding exon 18 of the NF1 gene, results from a deletion of 14 nucleotides at nucleotide positions 2109 to 2122, causing a translational frameshift with a predicted alternate stop codon (p.V705Ffs*6). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This alteration has been observed in at least one individual with a personal and/or family history that is consistent with NF1-related disease (Ambry internal data).
