Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006231.4(POLE):c.2101G>A (p.Glu701Lys), citing Ambry Variant Classification Scheme 2023: The p.E701K variant (also known as c.2101G>A), located in coding exon 19 of the POLE gene, results from a G to A substitution at nucleotide position 2101. The glutamic acid at codon 701 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
