Uncertain significance for Cardiovascular phenotype; Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_006767.4(LZTR1):c.2100G>T (p.Met700Ile), citing Ambry Variant Classification Scheme 2023: The p.M700I variant (also known as c.2100G>T), located in coding exon 18 of the LZTR1 gene, results from a G to T substitution at nucleotide position 2100. The methionine at codon 700 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
