Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000548.5(TSC2):c.1077C>A (p.Asp359Glu), citing Ambry Variant Classification Scheme 2023: The p.D359E variant (also known as c.1077C>A), located in coding exon 10 of the TSC2 gene, results from a C to A substitution at nucleotide position 1077. The aspartic acid at codon 359 is replaced by glutamic acid, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
